What is Tamoxifen?
Tamoxifen is a selective estrogen receptor modulator first made at Imperial Chemical Industries in the 1960s. It was approved in the US in 1977 for breast cancer and is still one of the most used cancer drugs in the world. This liquid uses the citrate salt.
A SERM blocks the estrogen receptor in some tissues and activates it in others. Tamoxifen blocks it in breast tissue but acts like estrogen in bone and the uterus.
How Tamoxifen works
Tamoxifen competes with estradiol for the estrogen receptor. When it binds, the receptor takes a shape that recruits co-repressors in breast cells, so estrogen-driven genes stay off.
In other tissues the same complex recruits co-activators instead, which is why it supports bone density and stimulates the uterine lining. In the hypothalamus and pituitary it blocks estrogen feedback, which raises LH and FSH.

What the research says about Tamoxifen
The NSABP P-1 trial, published by Fisher and colleagues in the Journal of the National Cancer Institute in 1998, reported about 49% fewer invasive breast cancers in high-risk women taking tamoxifen than on placebo.
Much of its activity comes from metabolites. The liver enzyme CYP2D6 turns tamoxifen into endoxifen, which binds the estrogen receptor far more strongly than the parent drug.
