Liothyronine is synthetic T3, triiodothyronine, the active form of thyroid hormone. The thyroid makes mostly T4, and tissues convert it to T3, which does most of the work inside cells.
It’s approved as Cytomel for hypothyroidism. In the lab it’s the standard way to switch on thyroid hormone receptors directly, without depending on the conversion step.
How T3 works
T3 enters the cell and binds thyroid hormone receptors (TR-alpha and TR-beta) in the nucleus. The receptor is already sitting on DNA, and T3 binding swaps its co-repressors for co-activators, so target genes switch on.
Those genes raise oxygen use, mitochondrial activity, heart rate and the breakdown of fat and protein. T3 also feeds back on the pituitary to lower TSH, which slows the thyroid’s own output.

What the research says about T3
T3 binds thyroid hormone receptors about ten times more tightly than T4, which is why it’s treated as the active hormone. Its half-life in blood is about one day, against roughly a week for T4.
Research uses it to study metabolic rate, heart rate, cholesterol handling and brain development, and in combination therapy trials that compare T3 plus T4 with T4 alone.
